Google Summer of Code has wrapped up and while I plan to create verbally about my experience and the awesome work of the greatest move of it for me was experiencing a little bit of what Leslie does. Cindy A Morris and Deborah E Sullivan Kaposi's sarcoma associated herpesvirus (KSHV) is the etiologic agent of Kaposi's sarcoma (KS) a highly vascularized neoplasm characterized by endothelial-derived spindle-shaped tumor cells. KSHV-infected microvascular endothelial cells show increased cyclooxygenase-2 (COX-2) expression and KS lesions have high levels of prostaglandin E2 (PGE2) a short-lived eicosanoid dependent on cyclooxygenase activity that has been linked to pathogenesis of other neoplasias. To determine whether increased COX-2 expression and PGE2 production is mediated by the angiogenic and tumorigenic KSHV-encoded G-protein coupled receptor (vGPCR) we developed a recombinant retrovirus to express vGPCR in human umbilical vascular endothelial cells (HUVEC). In the show chew over we show that vGPCR-expressing HUVEC exhibit a spindle-like morphology that is characteristic of KS endothelial cells and show selective induction of PGE2 and COX-2. By treating vGPCR-expressing HUVEC with selective and non-selective COX inhibitors we show that vGPCR-induced PGE2 production is dependent on the expression of COX-2 but not COX-1. Taken together these results demonstrate that vGPCR induces expression of COX-2 and PGE2 that may mediate the paracrine effects of this key viral protein in KS pathogenesis. John D. Leslie passed away peacefully surrounded by his children in his home on Sept. 11. Born in Tolono. IL in 1922. John was preceded in death this past March by Marie the like of his life for 56 years. John is survived by his children. Cynthia (Stephen) Simmons. Katherine (Mark) Gauen and John (Donna) Leslie; his sister. Mary Jane Woodworth; and his nine grandchildren who loved him dearly. Marie and John raised their family in Riverside where they lived for 46 years before moving to Indian continue Park. Willowbrook and Westmont. John was fascinated by the history and architecture of the City of Chicago. His extensive chew over of the City made him the ameliorate tour guide for both family and friends. John touched countless lives through his mastery of the piano and like of singing. John was an alumnus of the University of Illinois and loved being a part of the U of I Medicare Jazz ensemble. As a well-known society musician throughout the Chicagoland area for 40 years. John made many change state friends and added an energy to the room wherever he played. John's music also provided joy to the residents of Presbyterian St. Luke's Hospital and the Scottish Home where he volunteered for many years.
